Lymphotoxin-beta, a novel member of the TNF family that forms a heteromeric complex with lymphotoxin on the cell surface
Article Abstract:
The role of lymphotoxin-alpha (LT-alpha) in inducing inflammatory reactions was investigated. LT is related to the lymphokine tumor necrosis factor (TNF), which is well-known to induce inflammatory reactions. The study protocol involved the cDNA cloning of LT-beta, a 33-kilodalton protein which forms a complex with LT-alpha on the surface of activated T, B and LAK cells. The results showed that LT-beta is a type II membrane protein with significant homology to TNF, LT-alpha and the CD40 receptor ligand. These results indicate a role for LT-alpha-LT-beta complex in an inflammatory reaction distinct from that mediated by TNF.

Cytotoxicity by tumor necrosis factor is mediated by both p55 and p70 receptors
Article Abstract:
The suggestion that tumor necrosis factor cytoxicity is not mediated by the p70 receptor is refuted. NIH 3T3 cells and clonal analysis of C7 3T3 cells, from which three independent p70-overexpressing clones were produced showed that p55 and p70 receptors always interact to produce cell death. Results have shown that the p70 receptor promote the cytotoxic effects of TNF. Thus, the role of p70 receptor in mediating cytotoxicity can not be entirely ruled out.

A review of the latest data on the role of JuNKie (JNK) in the regulation of tumor necrosis factor (TNF) - dependent apoptosis is presented. A discussion on the current available knowledge about the decision taken by the cell to survive or die in response to tumor necrosis factor-alpha (TNF), which plays a pivotal role in orchestrating innate inflammatory responses in vertebrates, is also presented.
