Development of improved versatile broad-host-range vectors for use in methylotrophs and other gram-negative bacteria
Article Abstract:
Results indicate that the improved broad-host range vectors are versatile in that they can be maintained in a wide variety of gram-negative bacteria. Their use in the Methylobacterium extorquens AM1 enables studying the genetic regulation of the bacterium's metabolism and physiology.

Molecular and mutational analysis of a DNA region separating two methylotrophy gene clusters in Methylobacterium extorquens AM1
Article Abstract:
The DNA region containing the methylotrophy gene clusters of Methylobacterium extorquens AM1 was analyzed to characterize the genetic mechanisms that mediate one-carbon (C1) metabolism. Analysis of the 14.2 kb region containing the serine cycle genes, pqq genes and regulatory methanol oxidation genes of Methylobacterium extorquens AM1 indicated the presence of 15 open reading frames that mediate the synthesis of malyl-CoA-lyase. Furthermore, the DNA region contained three genes that encode enzymes for folate metabolism.

Molecular analysis of mxbD and mxbM, a putative sensor-regulator pair required for oxidation of methanol in Methylobacterium extorquens AM1
Article Abstract:
The nucleotide sequence of the genes encoding the MxbD and MxbM proteins of Methylobacterium extorquens AM1 were analyzed to characterize their expression and molecular characteristics. Nucleotide sequence analysis of the mxbDM region indicated the role of the genes on the regulation of the mxaF methanol dehydrogenase. Furthermore, the mxbD and mxbM genes encoded a sensor-regulator pair that mediate the regulation of methanol oxidation.
