Overexpression of the calcineurin target CRZ1 provides freeze tolerance and enhances the fermentative capacity of baker's yeast
Article Abstract:
The role of the salt-responsive calcineurin-Crz1p-pathway in mediating tolerance to freezing by industrial baker's yeast was investigated. It was found that the overexpression of CRZ1 in the industrial strain increased both salt and freeze tolerance improving the leavening ability of baker's yeast in high sugar dough suggesting that expression of calcineurin-Crz1p target genes can alleviate the harmful effects of ionic stress during freezing.

Water transport in intact yeast cells as assessed by fluorescence self-quenching
Article Abstract:
The fluorescence self-quenching method is used to assess the water transport in intact yeast cells loaded with 5- and-6-carboxyfluorescein. The results are similar to those obtained for protoplasts, which are assessed by using either fluorescence or light scattering.

A novel approach that combines yeast cDNA overexpression libraries with yeast surface display to allow the rapid flow cytometric screening of engineered yeast for gene products that improve the display of heterologous proteins is reported.
