Power analysis for real-time PCR quantification of genes in activated sludge and analysis of the variability introduced by DNA extraction
Article Abstract:
A study is conducted to determine the power of discrimination of the real-time PCR assay for monitoring fluctuations in microbial populations within activated sludge and to identify sample processing points where methodological changes are needed to minimize the variability in target quantification. Power analysis of real-time PCR assays indicated that three to five samples were necessary to detect a twofold increase in bacterial 16S rRNA genes and other samples to detect a fivefold increase in Nitrospira 16S rRNA genes.

Reduction of hexavalent uranium from organic complexes by sulfate- and iron-reducing bacteria
Article Abstract:
The effect of organic complexation of hexavalent uranium [U(VI)] on U(VI) reduction by Desulfovibrio desulfuricans, a sulfate-reducing strain, and Shewanella alga, an iron-reducing strain, is investigated. The nature and amount of organic matter complexed with U(VI) as well as the type of bacteria present influenced the reduction trend. The amount of U(VI) complex in solution was also shown to be inversely correlated with the rate of reduction.

Emergence of competitive dominant ammonia-oxidizing bacterial populations in a full-scale industrial wastewater treatment plant
Article Abstract:
Ammonia-oxidizing bacterial populations in an industrial wastewater treatment plant were investigated with amoA and 16S rRNA gene real-time PCR assays. The combination of real-time PCR assays for the AOB16S rRNA gene and the amoA gene allowed the detection of a change in competitive dominance between a cultured representative and an uncultured representative of AOB in the Nitrosomonas communis lineage.
