Probing activated sludge with oligonucleotides specific for proteobacteria: inadequacy of culture-dependent methods for describing microbial community structure
Article Abstract:
Oligonucleotide probes were used for analysis of microbial community structure in activated sludge. Group-specific cell counts were determined by in situ hybridization with fluorescent rRNA-targeted oligonucleotide probes. The activated sludge samples were dominated by proteobacteria from the alpha, beta or gamma subclass. Cultivation-dependent shifts in community composition was demonstrated.

In situ characterization of Nitrospira-like nitrite-oxidizing bacteria active in wastewater treatment plants

Researchers studied the activity of Nitrospira in wastewater treatment plants using 16S rRNA gene sequence analysis, fluorescence in situ hybridization, confocal laser scanning microscopy, and computer-aided three-dimensional visualization. Nitrospira-like bacteria are the predominant nitrite oxidizers in wastewater treatment plants.

Cultivation-independent, semiautomatic determination of absolute bacterial cell numbers in environmental samples by fluorescence in situ hybridization
Article Abstract:
Researchers used a combination of fluorescence in situ hybridization (FISH), confocal laser scanning microscopy, and digital image analysis to measure the number of ammonia-oxidizing bacteria in municipal nitrifying activated sludge. This is the first known use of this technique to quantify bacterial cells in aggregates or biofilms.
