Purification and characterization of the extracellular alpha-amylase from Streptococcus bovis JB1
Article Abstract:
The purification and characterization of the extracellular alpha-amylase produced by Streptococcus bovis JB1 were described. The purified alpha-amylase had an isoelectric point of 4.5 and an apparent molecular mass of 77 kDa, as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. It showed optimal activity from pH 5.0 to 6.0 and was stable to temperatures below 50 degrees C. The K(sub m) and k(sub cat) of the enzyme for soluble potato starch was 0.88 mg . ml-1 and 2,510 U . mg of protein-1, respectively. Amylose was the best substrate, with maltose, maltotriose and maltotetraose as the major end products.

Determining sources of fecal pollution in a rural Virginia watershed with antibiotic resistance patterns in fecal streptococci
Article Abstract:
A study to determine sources of fecal pollution in a rural Virginia watershed has been carried out using antibiotic-resistance patterns in fecal streptococci. Patterns of antibiotic resistance were analyzed by discriminant and cluster analysis. Besides finding the sources, the project was intended to validate the method using a larger database of known-source isolates from a wider geographical region than had been used before. Results validated use of the method and showed that water-quality improvement is possibly when efforts to deal with the identified sources and change the situation are made.

Comparison of C18-carboxypropylbetaine and glass bead DNA extraction methods for detection of Mycobacterium bovis in bovine milk samples and analysis of samples by PCR
Article Abstract:
The performance of the C18-carboxypropylbetaine (CB-18) and the glass bead DNA extraction methods in detecting Mycobacterium bovis in bovine milk samples by polymerase chain reaction has been evaluated. Both procedures were used on samples from 17 skin test-positive cattle. Results show that PCR for CB-18 was 94.1% and 58.8% for the glass bead approach. This finding indicates that CB-18 is the better method for the detection of M. bovis.
