Real-time observation of RecA filament dynamics with single monomer resolution
Article Abstract:
Single-molecule fluorescence assays and hidden Markov modeling are used to show the most direct evidence that a RecA filament grows and shrinks primarily one monomer at a time and only at the extremities. Results support the proposal that a sufficient role for the RecA-loading machineries in helping RecA replace single-stranded deoxyribonucleic acid (DNA) binding protein (SSB) is to provide a nucleation cluster-like structure to initiate RecA-filament extension.

Human Rad51 protein promotes ATP-dependent homologous pairing and strand transfer reactions in vitro
Article Abstract:
Human Rad51, a structural homolog of Escherichia coli RecA, mediates homology recognition and initiates strand exchange. However, additional proteins are needed in extensive heteroduplex formation in higher organisms. This was gleaned from an investigation of the biochemical properties of hRad51 in vitro. It was also found that the binding of hRad51 to dsDNA facilitates end-to-end joining by DNA ligase.

The roles of murine Pot1 homologs are examined and the way in which the cells protect their chromosome ends from DNA-repair activities are described. The computational analyses of transcript sequences and microarray profiling of splicing have provided a detailed understanding of the extent and complexity of AS in different cells and tissues.
