Requisite role of angiopoietin-1, a ligand for the TIE2 receptor, during embryonic angiogenesis
Article Abstract:
The role of the TIE2 receptor tyrosine kinase ligand, Angiopoietin-1, in the development of the vascular nervous system was analyzed in mice without Angiopoietin-1 expression. The Angiopoietin-1 null mutants exhibited defective hearts characterized by collapsed endothelial lining and underdeveloped ventricular endocardium. Furthermore, mutant mice without TIE2 receptors exhibited embryonic lethality during the first and second days of embryonic development due to deletions of the Angiopoietin-1 gene.

The receptor tyrosine kinase MuSK is required for neuromuscular junction formation in vivo
Article Abstract:
Agrin activates the MuSK (muscle-specific kinase) receptor to initiate the formation of the neuromuscular junction by signaling cascades. Agrin also activates presynaptic differentiation and synapse-specific transcription through MuSK receptor stimulation. MuSK gene disruption in mice resulted in perinatal death due to the aberrant formation of neuromuscular synapses. These results indicate the importance of MuSK as a signaling component for receptors that respond to neural agrin.

Mice lacking the CNTF receptor, unlike mice lacking CNTF, exhibit profound motor neuron deficits at birth
Article Abstract:
Mice containing mutated genes encoding the ciliary neurotrophic factor (CNTF) showed no noticeable defects or abnormalities in the nervous system. However, mice lacking the CNTF receptor displayed severe motor neuron abnormalities and died perinatally, indicating that CNTF receptors are essential for the normal development of the nervous system. Evidence also suggested that CNTF receptors serve as a receptor for a second CNTF-like ligand during neurotrophic activity.
