Reverse transcriptase (RT) inhibition of PCR at low concentrations of template and its implications for quantitative RT-PCR
Article Abstract:
The reverse transcriptase/polymerase chain reaction (RT-PCR) process is examined with particular attention to the common observation that control DNA reaction mixtures with RT do not provide DNA amplification during PCR while control DNA reaction mixtures without RT do not inhibit PCR. The findings indicate that the RT interaction with the specific mRNA or cDNA template mediates RT inhibition of the PCR and that such inhibitory effect is influenced by template concentration.

Analysis of nifH gene pool complexity in soil and litter at a Douglas fir forest site in the Oregon Cascade Mountain Range
Article Abstract:
Research was conducted to examine the nitrogen-fixing microbial communities in a Douglas fir forest on the western slope of the Oregon Cascade Mountain range. Restriction fragment polymorphism analysis followed a protocol for general and selective PCR amplification of nifH gene fragments. Results showed insights into the distinctness of the nitrogen-fixing microbial communities in litter and soil in a Douglas fir forest.

Application of reverse transcriptase PCR for monitoring expression of the catabolic dmpN gene in a phenol-degrading sequencing batch reactor
Article Abstract:
A reverse transcriptase polymerase chain reaction (RT-PCR) batch sequencer coupled with freeze-thaw extraction method indicate that synthesis and lifetime of mRNA coded by dmpN gene are enhanced by increase in concentration of phenol. The dmpN gene in Pseudomonas putida ATCC 11172 codes for phenol hydroxylase. This organism can be used to degrade phenol in the environment which is released from industrial processes.
