Selection of clc, cba, and fcb chlorobenzoate-catabolic genotypes from groundwater and surface waters adjacent to the Hyde Park, Niagara Falls, chemical landfill
Article Abstract:
The frequency of isolation of the nonhomologous chlorobenzoate catabolic genotypes clc, cba and fcb has been investigated. To this end, 464 samples were collected from both contaminated and non-contaminated industrial landfill sites in the Niagara watershed at Hyde Park during spring, summer and fall. Results show that the genotypes were found in less than 10% of the isolates from the contaminated sites, suggesting that a high level of genetic diversity exists in the microbial community. The known genes were not enriched from the adjacent noncontaminated control sites.

Involvement of a chlorobenzoate-catabolic transposon, Tn5271, in community adaptation to chlorobophenyl, chloroaniline, and 2,4-dichlorophenoxyacetic acid in a freshwaster ecosystem
Article Abstract:
The role of chlorobenzoate-catabolic transposon, Tn5271, in community adaptation was investigated. The transposon was inserted into the conjugative plasmid (pBR60) of the natural host Alcaligenes sp. strain BR60. Then, the populations of the host and the organism carrying Tn5271 was determined after exposure to different chemical environment. Results showed that Tn5271 allowed the utilization of 3-chlorobenzene, 3-chlorobiphenyl but played no role in the degradation of 2,4-dichlorophenoxyacetate.

The nucleotide sequence of the Tn5271 3-chlorobenzoate 3,4-dioxygenase genes (cbaAB) unites the class IA oxygenases in a single lineage
Article Abstract:
Sequence analysis helps determine the nucleotide sequence of the 3-chlorobenzoate 3,4-dioxygenase genes (cbaAB) of the transposon Tn5271. Mutagenesis enables the evaluation of the activity of two sequenced open reading frames. Reductase and dioxygenase proteins are encoded by the cbaB and cbaA genes, respectively. A comparative study of the sequences of the cbaAB genes with those of other oxygenases reveals a single lineage among the class IA oxygenases.
