Substrate specificity of and product formation by muconate cycloisomerases: an analysis of wild-type enzymes and engineered variants
Article Abstract:
Research was conducted to investigate the substrate specificity of the muconate cycloisomerases from Pseudomonas putida PRS2000 and Acinetobacter "calcoacetus" ADP1. Kinetic improvements in the variants of P. putida muconate cycloisomerase were achieved by site-directed mutagenesis but was not accompanied by a change from protoanemonin to cis,cis-dienelactone as the product of 3-chloro-cis,cis-muconate conversion. Results suggest that the different catalytic features of muconate and chloromuconate cycloisomerases are caused by a number of changeable characteristics.

Export of cytochrome P450 105D1 to the periplasmic space of Escherichia coli

Researchers have expressed cytochrome P450 105D1 from Streptomyces griseus in E. coli, where it is exported to the periplasmic space. This cytochrome has a very broad substrate range, including drugs, agrochemicals, and environmental pollutants.

Evaluation of p-naphtholbenzein-beta-D-galactoside as a substrate for bacterial beta-galactosidase

Researchers have created a new substrate for detecting microbial beta-galactosidase that turns bacterial colonies on agar pink. The substrate was hydrolyzed by over 90% of the 206 Enterobacteriaceae strains tested.
