Tethering RITS to a nascent transcript initiates RNAi- and heterochromatin-dependent gene silencing
Article Abstract:
The tethering of the RNA-Induced Transcriptional Silencing (RITS) subunit, Tas3, to the RNA transcript of the normally active ura[4.sup.+] gene that silences ura[4.sup.+] expression is demonstrated. Results reveal that this silencing depends on a functional RNAi pathway, requires the heterochromatin proteins and is accompanied by the generation of ura[4.sup.+] siRNAs, histone H3-lysine 9 methylation and Swi6 binding.

A long-distance relationship between RNAi and Polycomb
Article Abstract:
RNA interference (RNAi) pathways can result in sequence-specific transcriptional gene silencing on the level of chromatin. It is shown that the RNAi machinery is required for long-distance physical interactions between chromosomes mediated by the Polycomb repressive complex and these results suggest that the RNAi machinery may regulate higher-order nuclear organization.

The role of the RNAi machinery in heterochromatin formation
Article Abstract:
The RNAi machinery is associated not only with target RNA translation but also with the formation of heterochromatin. Evidences show that RNA-mediated chromatin modifications are found to play an important role in epigenetic transcriptional genes silencing.
