Spontaneous rDNA copy number variation modulates Sir2 levels and epigenetic gene silencing
Article Abstract:
A study shows that in budding yeast large ribosomal DNA (rDNA) deletions arise frequently and cause an increase in telomeric and mating-type gene silencing proportional to repeat loss. The evidence for a reduction in the fraction of Sir2 colocalizing with the nucleolar marker Nop1, and for SIR2 autoregulation is presented.

The silencing of euchromatic genes is achieved by an interaction between DNA methyltransferase 1 (DNMT 1) and three members of Heterochromatin Protein 1family. The methylation of DNA and histones are found to be interdependent.

Functional cooperation between HP1 and DNMT1 mediates gene silencing

The interaction between histones and DNA methyltransferases is responsible for the silencing of mammalian euchromatic genes. The Heterchromatic Protein 1 family members mediate the methylation of DNA and histones.
