Thaumatin production in Aspergillus awamori by use of expression casettes with strong fungal promoters and high gene dosage
Article Abstract:
The sweet protein thaumatin II has been overexpressed in Aspergillus awamori lpr66 by using four expression cassettes with strong fungal promoters, a signal sequence for protein translocation, a KEX protease cleavage site and the synthetic gene (tha) which encodes thaumatin II. Aspergillus awamori lacks the PepA protease while thaumatin was originally extracted from the arils of the fruit of Thaumatococcus danielli Benth. It is used as food sweetener and to enhance the palatability of animal feed. The best expression results were obtained with the gdhA promoter of A. awamori or with the gpdA promoter of Aspergillus nidulans.

Article Abstract:
Promoters of the cephamycin gene cluster in Nocardia lactamdurans are studied through the synthesis of promoter probe vectors having the promoterless amy gene of Streptomyces griseus. These promoters bind to the positively acting regulatory protein CcaR to induce cephamycin biosynthesis in N. lactamdurans. Amplification of the CcaR gene, however, leads to the oversecretion of cephamycin. The -10 and upstream regulatory regions are easily recognized by the RNA polymerase.

Substrate analysis and molecular cloning of the extracellular alkaline phosphatase of Streptomyces griseus
Article Abstract:
Streptomyces griseus and the substrate analysis and molecular cloning of its extracellular alkaline phosphatase (AP) are discussed. The pure enzyme was found to use various organic phosphorylated compounds as substrates. It is different from the phosphatases of Gram-negative bacteria.
