Transposon mutagenesis with IS6100 in the avermectin-producer Streptomyces avermitilis
Article Abstract:
Research was conducted to evaluate the intermolecular transposition of the insertion sequence IS6100 from the delivery plasmid to the genome of the avermectin-producer Streptomyces avermitilis. Results revealed the consistency of Southern hybridization with random transposition. Variances involving the frequency of reversion of different auxotrophic mutations by precise excision were also observed.

Article Abstract:
Disruption of sblA in Streptomyces lividans is discussed. This disruption makes expression of a heterologous alpha-amylase gene in the presence of glucose possible. In a transposition mutant of S. lividans TK24, the usually glucose-repressible expression of a heterologous alpha amylase gene was rendered resistant to glucose repression.
