Twenty-five years of the nucleosome, fundamental particle of the eukaryote chromosome
Article Abstract:
A study made in 1974 suggesting the that the chromatin structure is based on a repeating unit of eight histone molecules and some 200 DNA pairs served as a basis for subsequent studies on the repeating unit, called nucleosome. One of these studies is about micrococcal nuclease digestion of chromatin, which allowed for the determination of the structure of the nucleosome. Another study led to the discovery of histone acetyl-transferase activity of transcription factors, which paved way for understanding the regulatory role of the nucleosome.

Histone octamer transfer by a chromatin-remodeling complex
Article Abstract:
The interaction of exogenous free DNA fragments with the activated chromatin-remodeling complex (RSC)-nucleosome complex was investigated to examine the histone-DNA contacts in the activated state. Results showed that the RSC serves as a catalyst to transfer a histone octamer from a nucleosome core particle to naked DNA. The reaction was also found to require an adenosine triphosphate and involve an activated RSC-nucleosome intermediate.

Activated RSC-nucleosome complex and persistently altered form of the nucleosome
Article Abstract:
Research was conducted to examine the process involving the creation of a discrete RSC-nucleosome complex, its transformation into an activated state via adenosine triphosphate analysis and the subsequent release of a revised nucleosome. The complex features a full complement of histones and is susceptible to endo- and exo-nucleases. Results revealed the existence of three states of nucleosomal configuration.
