Use of 4-nitrophenoxyacetic acid for detection and quantification of 2,4-dichlorophenoxyacetic acid (2,4-D)/alpha-ketoglutarate dioxygenase activity in 2,4-D-degrading microorganisms
Article Abstract:
The 2,4-dichlorophenoxyacetic acid (2,4-D)/alpha-ketoglutarate dioxygenase (TfdA) of Alcaligenes eutrophus was analyzed by spectrophotometric assay with 4-nitrophenoxyacetic acid (4-NPAA). Spectrophometric analysis of the 2,4D-catalyzing enzymes of Alcaligenes eutrophus produced negative results for 4-NPAA degradation and exhibited no activity in the extracts. Furthermore, the TfdA isolates from Alcaligenes eutrophus did not exhibit color production in selected whole cells due to the inability of these cultures to transport 4-NPAA.

Existence of a hydrogen-sulfide:ferric ion oxidoreductase in iron-oxidizing bacteria
Article Abstract:
The presence of a hydrogen sulfide:ferric ion oxidoreductase (SFORase) in different iron-oxidizing bacteria was discussed. In this study, SFORase activity in intact cells and cell extracts of various strains of Thiobacillus ferrooxidans and Leptospirillum ferrooxidans was assayed. Results showed that the SFRoase activity in all the iron-oxidzing bacteria tested was comparable tp that of T. ferrooxidans AP19-3. These indicated the existence of SFORase in diffenet iron-oxidizing bacteria.

Purification and characterization of gallic acid decarboxylase from Pantoea agglomerans T71
Article Abstract:
A study was conducted to characterize and purify gallic acid decarboxylases from Pantoea agglomerans T71. Strain T71 was determined from a soil sample from an orchard in Okayama, Japan. Decarboxylase activity was assayed at 30 degrees C in a 2 ml buffer solution while the Shimadzu model LC-6A HPLC system was utilized to analyze gallic acid and pyrogallot. Experimental results indicated that gallic acid decarboxylase is sensitive to oxygen.
