Wnt-4 expression induces a pregnancy-like growth pattern in reconstituted mammary glands in virgin mice
Article Abstract:
Expression of Wnt-1 in the mammary gland may activate a developmental pathway that is normally controlled by the related gene Wnt-4. A recombinant retrovirus was used to express the Wnt-4 gene in transplanted mammary epithelium grown in virgin mice. Wnt-4 may control branching of the epithelium in early pregnancy. Expression of Wnt-4 in fully grown glands produced ducts that had more branches than normal and stimulated premature development of alveoli. The changes associated with Wnt-4 expression resemble those produced by Wnt-1, a gene that is not normally expressed in the mammary gland, but is activated by mouse mammary tumor virus.

Ectopic TGF-beta-1 expression in the secretory mammary epithelium induces early senescence of the epithelial stem cell population
Article Abstract:
Analysis of the ectopic expression of a transforming growth factor-beta-1 (TGF-beta-1) influenced by a whey-acidic protein promoter (WAP) indicates that this expression affects lobular progenitors and induces the mammary ductal epithelium to age rapidly, thereby inhibiting lactation and generation of secretory mammary epithelium through an intracrine mechanism. These studies reveal the presence of two different progenitor cells, each involved in lobular and ductal functions.

Developmental regulation of alpha-fetoprotein expression in intestinal epithelial cells of transgenic mice
Article Abstract:
The alpha-fetoprotein (AFP) repressor is only active in the goblet cells of the intestine in mice. The repressor element is located between -838 and -259 bp upstream from the AFP gene. Deletion of the repressor causes continued expression of AFP in goblet cells and enteroendocrine cells, but not in enterocytes.
