Efficient gene silencing with high selectivity using mission shRNA lentiviral vectors
Article Abstract:
RNA interface technology is one of the key biological breakthroughs and has revolutionalised basic biology and gene function studies but it also holds promise to revolutionalise drug discovery and therapeutics. The MISSION shRNA libraries overcome the limitations of synthetic small interface RNAs (siRNAs) by providing a system for long term silencing and phenotypic observations and the ability to generate lentiviral particles for infection and integration of primary cells, dividing cells and non dividing cells.

The RNA-induced initiation of transcriptional gene silencing (RITS) complex is required for heterochromatic gene silencing, but it also contains short interference (si)RNAs that are generated by dicer (Dcr), an enzyme involved in RNAi. RITS localizes at the specific loci and mediates the processing of nascent RNA transcription by recruiting Dcr or RNA-dependent RNA polymerase and generates more siRNA and continuing the process.

The RNase III dicer enzyme processes RNAs into small (si)RNAs and micro (mi)RNAs that function as guides for RNA-induced silencing complex (RISC) to cleavage messenger RNA (si)RNAs or block its translation (mi)RNAs. The dicer-1 (Dcr-1) and dicer-2 (Dcr-2) have distinct but overlapping role in the siRNA and miRNA silencing pathways and they might direct the assembly of functionally different RISCs.
