Hematopoietic stem cell gene transfer in a tumor-prone mouse model uncovers low genotoxicity of lentiviral vector integration
Article Abstract:
The hematopoietic stem cells from the Cdkn[2a.sup.-/-] tumor-prone mice to assess the oncogenicity of the prototypical retroviral and lentiviral vectors are exploited and then transduced them in matched clinically relevant conditions, and compared integration site selection and tumor development in transplanted mice. The results validate the much-needed platform to investigate vector safety and proved that the prototypical lentiviral vectors have low oncogenic potential, highlighting a major rationale for application to gene therapy.

Coordinate dual-gene transgenesis by lentiviral vectors carrying synthetic bidirectional promoters
Article Abstract:
Synthetic bidirectional promoters that mediate coordinate transcription of two mRNAs in a ubiquitous or tissue-specific manner was developed based on the finding that some cellular promoters intrinsically promote divergent transcription. After direct delivery or transgenesis, lentiviral vectors incorporating the new promoters enabled efficient dual gene transfer in several tissues.

Endogenous microRNA can be broadly exploited to regulate transgene expression according to tissue, lineage and differentiation state

An analysis conducted to study the potential of the theory that transgene expression can be suppressed in hemotopoietic cells using vectors that are responsive to microRNA (miRNA) regulations for more complex control of transgene expression. It is found that a threshold miRNA concentration is needed for transgene suppression.
