Mammalian cell-based optimization of the biarsenical-binding tetracysteine motif for improved fluorescence and affinity
Article Abstract:
Residues surrounding the tetracysteine motif are randomized and fused to GFP, retrovirally transduced into mammalian cells and iteratively sorted by fluorescence-activated cell sorting for high FRET from GFP to ReAsH in the presence of increasing concentrations of dithiol competitors. The selected sequences reveal higher fluorescence quantum yields and markedly improved dithiol resistance, culminating in a >20-fold increase in contrast.

Article Abstract:
A protein changes its behavior when it is modified with a thiol-specific probe, thus complicating the task of determining the fraction of the protein that is modified. Researchers have circumvented these difficulties with the used of general method for visualizing thiol modifications that allows accurate and simultaneous quantification of the in vivo thiol-disulfide status of hundreds of cellular proteins.

Blue fluorescent proteins with enhanced brightness and photostability from a structurally targeted library
Article Abstract:
The blue fluorescent protein (BFP), new Azurite, is well expressed in bacterial and mammalian cells and has extended the palette of fluorescent proteins that can be used for imaging. Brighter BFPs are valuable in multicolor fluorescents experiments and also as Forster resonance energy transfer (FRET) donors to green fluorescent protein (GFP) from Aequorea victoria.
