Enzyme activity to augment the characterization of tethered bilayer membranes
Article Abstract:
The rate of [Ca.sup.2+]-triggered phospholipase [A.sub.2] degradation of tethered bilayer membranes (tBLMs) composed of a synthetic liquid, beta-mercaptoethanol and palmitoyloleoylphosphatidylcholine is approximately 80 times greater than for those prepared with diphytanoylphosphatidylcholine. Electrochemical impedence spectroscopy and neutron reflectivity data indicate complete, water-free tBLMs that exhibit near ideal capacitive behavior and the presence of a water reservoir in the bilayer subspace proximal to the substrate surface for both tBLMs.

Interaction of the neurotransmitter, neuropeptide Y, with phospholipid membranes: infrared spectroscopic characterization at the air/water interface
Article Abstract:
External infrared reflection absorption spectroscopy is used to investigate the association of neuropeptide Y (NPY) at the air/water interface and with phospholipid monolayers on water as subphase. The peptide was found to adopt a conformation that is dominated by the [alpha]-helical motif at lipid monolayers, both charged and zwitterionic, below the peptide collapse pressure [[pi].sub.col], identical to secondary structure in neat peptide layers at aqueous surfaces.

The adsorption of neuropeptide Y (NPY) with air-water interfaces and with phospholipid monolayers on water subphases and on physiological buffer is investigated. Surface pressure versus molecular area solutions of the peptide at water surfaces depend on the concentration of the spreading solutions.
