11-color, 13-parameter flow cytometry: identification of human naive T cells by phenotype, function, and T-cell receptor diversity
Article Abstract:
A flow cytometry technique has been developed that precisely characterizes naive CD4 and CD8 cells by cytokine production, phenotype, and T-cell receptor diversity. This technique can identify at least 75 phenotypical subsets of lymphocytes in human blood, which in turn permits examination of unmodified antigen-reactive T cells from cancer patients.

Beyond six colors: a new era in flow cytometry
Article Abstract:
The history, application, and future trends in the field of flow cytometry are discussed. Recent improvements in cytometers enable independently detecting as many as 12 different molecules in the peripheral blood lyphocytes. Application of multicolor technology through the development of polychromatic flow cytometry has poised cytometry for precise identification of cellular subsets of lymphocyte populations.

Quantum dot semiconductor nanocrystals for immunophenotyping by polychromatic flow cytometry
Article Abstract:
Experiments were carried out to highlight the importance of multiparametric approaches to the phenotypic analysis of T cells to show the utility and broad applicability of quantum dots in the ongoing development of polychromatic flow cytometry. It was found that quantum dots could be used to produce bright and highly specific multimeric probes for antigen-specific T cells (comparable to standard pMHC tetramers), and that these pMHCI-quantum dot complexes might be prepared in a wide variety of colors.
