Amelioration of inflammatory arthritis by targeting the pre-ligand assembly domain of tumor necrosis factor receptors
Article Abstract:
A hypothesis that pre-ligand binding assembly domain (PLAD)-mediated blockade of tumor necrosis factor receptor (TNFR) assembly might inhibit TNF-alpha signaling and ameliorate TNF-induced disease in a receptor-specific fashion is presented. Results reveal that soluble versions of PLAD, especially those derived from P60, block the biochemical effects of TNF-alpha in vitro and potently inhibit arthritis in animal models.

Adiponectin protects against myocardial ischemia-reperfusion injury through AMPK- and COX-2--dependent mechanisms
Article Abstract:
The role of adiponectin in myocardial remodeling in response to acute injury is examined. Ischemia-reperfusion in adiponectin-deficient (APN-KO) mice resulted in increased myocardial infarct size, myocardial apoptosis and tumor necrosis factor (TNF)-alpha expression compared with wild-type mice.

Angiopoietin-2 sensitizes endothelial cells to TNF-alpha and has a crucial role in the induction of inflammation
Article Abstract:
The angiopoietins Ang-1 and Ang-2 are identified as ligands of the receptor tyrosine kinase Tie-2. Ang-2 is an autocrine regulator of endothelial cell inflammatory responses and acts as a switch of vascular responsiveness.
