Intractable diarrhoea associated with secondary amyloidosis in rheumatoid arthritis
Article Abstract:
Treatment-resistant diarrhea can be a serious complication in patients with rheumatoid arthritis (RA) and amyloidosis. The medical records of 24 patients with RA, confirmed amyloidosis, and treatment-resistant diarrhea lasting for a mean of 87 days were evaluated. Laboratory tests indicated poor intestinal protein absorption and high C reactive protein levels common in response to infection. A few patients developed intestinal blockages or colon inflammation due to impaired blood supply. Corticosteroid therapy was effective in many patients. Nearly 39% of the patients died within five years, half due to infection complications.

v-ATPase [V.sub.O] subunit d2-deficient mice exhibit impaired osteoclast fusion and increased bone formation
Article Abstract:
A study was conducted to show that inactivation of Atp6vOd2 in mice results in markedly increased bone mass due to defective osteoclasts and enhanced bone formation. The results identified Atp6vOd2 as a regulator of osteoclast fusion and bone formation and provide genetic data showing that it is possible to simultaneously inhibit osteoclast maturation and stimulate bone formation by therapeutically targeting the function of a single gene.

Osteoclasts, no longer osteoblast slaves
Article Abstract:
A study identified a protein Atp6v0d2, a component of a proton pump, which unexpectedly promotes precursor fusion and osteoclast activity but inhibits osteoblasts. The findings raise the possibility of therapeutic intervention for osteoporosis by targeting a single gene product in osteoclasts to inhibit bone destruction and stimulate bone formation.
