Periarticular osteopenia in adjuvant induced arthritis: role of interleukin-1 in decreased osteogenic and increased resorptive potential of bone marrow cells
Article Abstract:
Interleukin-1 (IL-1) may be involved in the bone degeneration seen in the arthritic process. Interleukin-1 is one of the mediators of the inflammatory process. Bone marrow cells have the capability to become bone-forming cells or bone-digesting cells. Arthritis was induced in rats by injecting killed Mycobacterium butyricum. After development of arthritis, bone marrow was harvested, the cells cultured, and a series of experiments carried out to evaluate the effect of arthritis and inflammatory substances, including IL-1, on the ability of marrow cells to development into bone forming or bone destroying cells. Bone marrow cells from arthritic rats showed increased potential to develop into bone destroying cells, a potential that was further increased in the presence of IL-1 and decreased when IL-1 activity was suppressed.

Lack of causal relationship between inducibility/severity of adjuvant arthritis in the rat and disease associated changes in production of nitric oxide by macrophages
Article Abstract:
The production of nitric oxide (NO) by macrophages does not appear to be related to the induction or severity of arthritis in rats. NO production in these white blood cells is increased as part of the inflammatory response. Rats from three different strains had tuberculosis bacteria injected into a foot to induce a type of autoimmune arthritis. The strains varied in their susceptibility to having arthritis induced. Macrophages were collected from the tissue lining the abdominal cavity, cultured, and the culture fluid was analyzed for NO. All rats from two of the strains developed arthritis, but some rats of the third strain did not. NO production increased markedly in all animals regardless of whether they developed arthritis. Moreover, the amount of NO did not correlate with the severity of arthritis.

Superinduction of interleukin 8 mRNA in activated monocyte derived macrophages from rheumatoid arthritis patients

The authors present research based on analysis using reverse transcriptase-polymerase chain reaction of macrophages derived from monocytes of rheumatoid arthritis patients showing that interleukin-8 is produced by the cells themselves as part of their function. This distinguishes its presence as not being a part of other circulation, and is relevant to understanding the function of leucocytes moving to the joints..
