Role of nucleotide cyclases in the inhibition of pregnant rat uterine contractions by the openers of potassium channels
Article Abstract:
Adenylate and guanylate cyclases are both involved in the opening of ATP-dependent potassium channels, whereas adenylate cyclase alone is involved in the opening of calcium-dependent potassium channels. The opening of potassium channels reduces the excitability of uterine smooth muscle.

Magnesium appears to stimulate the production of nitric oxide, which is a naturally occurring substance that relaxes blood vessels. This would explain why magnesium relaxes blood vessels and smooth muscle.

Roles of potassium channels and nitric oxide in modulation of uterine contractions in rat pregnancy
Article Abstract:
Calcium-dependent potassium channels appear to become less sensitive to nitric oxide as pregnancy progresses. Consequently, nitric oxide can no longer suppress uterine contractions, which is one of the steps leading to labor.
