Self-fusion of the ALL1 gene: a new genetic mechanism for acute leukemia
Article Abstract:
The ALL1 gene may be involved in twice as many cases of acute leukemia in adults as previously thought due to a gene defect called self-fusion. Self-fusion occurs when the internal portion of a gene is duplicated and fuses with itself. The result of ALL1 self-fusion is the separation of the amino-terminal region of the ALL1 protein from the rest of the protein. ALL1 self-fusion does not produce visible chromosomal rearrangements and can only be identified through molecular analyses. Researchers identified ALL1 gene arrangements in three of four patients who had acute myelogenous leukemia (AML) and a chromosomal abnormality called trisomy 11. In two of these cases, the rearrangements involved self-fusion of the ALL1 gene. Fusion of the ALL1 gene with genes on other chromosomes is involved in approximately 5% to 10% of acute leukemias. ALL1 gene self-fusion may account for an additional 5% of adult acute myelogenous leukemia cases.

Molecular basis of mature T-cell leukemia. (Special Communication)
Article Abstract:
Four genes appear to be involved in the development of T-cell leukemia. They are all located at the TCL1 locus on chromosome 14q32.1. The expression of these genes and their role in T cell leukemia is reviewed.
