Article Abstract:

Experimental microdialysis probes to measure A(beta) levels in brain interstitial fluid and local brain electrical activity have shown that increases in hippocampal electrical activity lead directly to rises in local level of interstitial fluid A(beta). Conversely, inhibiting neuronal or synaptic activity in vivo resulted in a substantial fall in A(beta) in a matter of hours, and it was confirmed that the process of synaptic vesicle exocytosis is associated with release of neuronal A(beta) into the extracellular fluid.

Amyloid beta protein immunotherapy neutralizes Abeta oligomers that disrupt synaptic plasticity in vivom

Article Abstract:

A study examines whether active immunization against pre-aggregated synthetic Abeta1-40/42 generates endogenous antibodies to Abeta that can prevent the inhibition of long-term potentiation (LTP) by natural Abeta oligomers in vivo. The ability of exogenous and endogenous antibodies to rapidly neutralize soluble Abeta oligomers that disrupt synaptic plasticity in vivo suggests that treatment with such antibodies might show reversible cognitive deficits in early Alzheimer disease.
