Upregulatory mechanisms compensate for mitochondrial DNA depletion in asymptomatic individuals receiving stavudine plus didnosine
Article Abstract:
The certainty that the homeostatic mechanisms are able to compensate for this mtDNA depletion in patients receiving stavudine plus didnosine(d4T+ ddI), an antiretroviral combination with great in vitro and in vivo capacity to decrease mtDNA is determined. The study concludes that treatment with ddT=ddI is associated with decreased mitochondrial mass and mtDNA content but that cytochrome c oxidase subnit II (COX-II) expression and COX activity remain unaltered.

The selection of the nucleoside/nucleotide reverse transcriptase inhibitor (NRTI/NtRTI) backbone is an important step in constructing a regimen for the treatment of HIV infection. Data published since 2000 that address NRTI/NtRTI backbone performances and safety when included in HAART regimen is reviewed.

Effect of nucleoside reverse transcriptase inhibitors on mitochondrial DNA synthesis in rats and humans
Article Abstract:
A reliable method for measuring mitochondrial DNA (mtDNA) synthesis has been developed and confirms that AZT reduces mtDNA synthesis and mitochondrial biogenesis in rats. Carefully designed clinical studies will be required to test this hypothesis definitively.
