Vav3 proto-oncogene deficiency leads to sympathetic hyperactivity and cardiovascular dysfunction
Article Abstract:
Studies related to sympathetic hyperactivity and cardiovascular dysfunction have identified Vav3, a GDP-GTP exchange factor that stimulates Rho and Rac GTPases, as an essential factor regulating the homeostasis of the cardiovascular system. Results of the study have uncovered the crucial role of Vav3 in the regulation of sympathetic nervous system (CNS) and cardiovascular physiology, and reveal a signaling pathway involving tachycardia and sympathetic hyperactivity with unknown etiologies.

The discovery and development of lovastatin, the first approved inhibitor of 3-hydroxy-3-methyl-glutaryl-CoA (HMG-CoA) reductase and the clinical trials that have provided the evidence for the ability of drugs to reduce the morbidity and mortality associated with cardiovascular disease are discussed. HMG-CoA reductase is the rate-limiting enzyme in the cholesterol biosynthetic pathway.

Endothelial cells, stimulated by the vascular shear stress and hypoxia further triggers the secretion of nitric oxide and hence results in vasodilatation in a human body. The analysis shows that the purinergic signaling is of great importance in curing the cardiovascular diseases, though the development of drugs targeting different subtypes is still awaited.
