Evidence of estrogen- and TCDD-like activities in crude and fractionated extracts of PM10 air particulate material using in vitro gene expression assays
Article Abstract:
A demonstration of the viability of in vitro gene expression assays in identifying endocrine disruptors was conducted using inhalable air particulate matter collected from an urban location. The particulate matter was made to react with human breast cancer cells and tested for endocrine disruption activity with Hepa 1c1c7 cells that were transfected with CYP1A1-regulated reporter gene. Results showed that polyaromatic hydrocarbons were responsible for the estrogenic and 2,3,7,8-tetrachlordibenzo-p-dioxin-like activity.

In vitro bioassays for assessing estrogenic substances
Article Abstract:
Accurate assessments of the potential estrogenic activity of chemicals or complex mixtures require the use of in vivo and in vitro assays. In vitro assays such as competitive ligand binding, cell proliferation and estrogen receptor transcription assays are currently being used to identify and assess chemicals that reportedly elicit estrogenic effects. The advantages and disadvantages of each of these assays are discussed.
