Crystal structure of nerve growth factor in complex with ligand-binding domain of the TrkA receptor
Article Abstract:
Many signalling processes, including cell differentiation and apoptosis of neurons, involve nerve growth factor (NGF), which binds to its two cell-surface receptors, p75 and TrkA. A high-affinity binding site for NGF is formed by the tyrosine kinase activity of TrkA. A new study investigates the crystal structure of human NGF in complex with human TrkA-d5 at high resolution. It is shown that the ligand-receptor interface comprises two similarly-sized patches. Results of the analysis are presented.

A structure based computational method that can drastically redesign protein ligand-binding specifications is presented. The biological and biosensing activities of the engineered receptors illustrate potential applications of computational design in controlling biomolecular recognisition.
