Feedback inhibition of calcineurin and Ras by a dual inhibitory protein Carabin
Article Abstract:
An endogenous inhibitor of calcineurin, named Carabin, is identified that also inhibited the Ras signalling pathway through its intrinsic Ras GTPase-activating protein (GAP) activity. The knockdown of Carabin by short interfering RNA has increased the production of interleukin-2 by antigen-specific T cells in vitro and in vivo, indicating that Carabin is a negative feedback inhibitor of the calcineurin signalling pathway, which also mediates crosstalk between calcineurin and Ras.

The neurodegenerative disease protein aprataxin resolves abortive DNA ligation intermediates
Article Abstract:
Purified aprataxin (APTX) protein and extracts derived from either APTX-defective chicken DT40 cells or Apt[x.sup.-/-] mouse primary neural cells were used to show that aprataxin resolves abortive DNA ligation intermediates. The result data indicate that neurological disorders associated with APTX mutations might be caused by the gradual accumulation of unrepaired DNA strand breaks resulting from abortive DNA ligation events.
