Mutations make enzyme polymerize
Article Abstract:
The formation of amyloid fibers from monomeric proteins has been linked with Alzheimer's disease and transmissible encephalopathies. A recent study by Pepys et al. provides insights into the properties and structures of two mutant human lysozymes that cause fatal amyloidoses. However, the structures do not clarify why amyloid fibers result from the mutant lysozymes. Another issue that needs clarification is the difference between the formation of amyloid fibres by mutant lysozymes and prion proteins.

Editing of ubiquitin conjugates by an isopeptidase in the 26S proteasome
Article Abstract:
A bovine isopeptidase that is appropriate for editing (poly)Ub-protein conjugates according to the degree of ubiquitination is analyzed. The enzyme features a unique localization and specificity. It is an intrinsic subunit of PA700, the 19S regulatory complex of the 26S proteasome. The isopeptidase can selectively salvage poorly ubiquitinated or slowly degraded Ub-protein conjugates from proteolysis by disassembling the degradation signal from the distal end of (poly)Ub chains.
