Presenilin is required for activity and nuclear access of Notch in Drosophila
Article Abstract:
Presenilin is required for the ligand-dependent nuclear access of the intracellular domain in Drosophila. However, it remains uncertain whether presenilin directly mediates proteolytic release of the intracellular domain or if it has a more indirect affect, possibly by activating a protease or mediating its transit to the plasma membrane. It has been established that null mutations in the Drosophila Presenilin gene eliminate Notch signal transduction and stop its intracellular domain entering the nucleus.

Article Abstract:
Cases of Alzheimer's disease involving autosomal dominant inheritance of mutations in the genes presenilin 1 (PS1) and presenilin 2 (PS2) are studied. PS1 mutations, a multipass transmembrane protein, were attributed to 25% of the cases involving familial Alzheimer's disease. This gene type is a functional homologue of a 'Caenorhabditis elegans' protein called SEL-12.
