Requirement for ceramide-initiated SAPK/JNK signalling in stress-induced apoptosis
Article Abstract:
Stress effects in U937 human monoblastic leukemia and bovine aortic endothelial cells were studied to determine the effect of relating the sphingomyelin and SAPK/JNK signalling systems in the induction of cell death or apoptosis. C2-ceramide was found to effect apoptosis as measured by DNA-specific fluorochrome bisbenzimide and confirmed by TUNEL assay. SAPK activation by C2-ceramide and sphingomyelinase was forty times than the control. Stress-induced apoptosis was reported to be initiated by ceramide but with an operational SAPK/JNK cascade as a requirement.

An SH2 domain in disguise
Article Abstract:
Cbl, a signalling protein from humans, contains an Src-homology-2 (SH2) domain that is so divergent in amino-acid sequence that scientists had not previously thought that it would be present in Cbl. The identification of this SH2 domain indicates that SH2 domains are more widely distributed in nature than their present count would suggest. Another very interesting observation is the presence in Cbl of a small helical domain situated just before the SH2 domain.

The biochemical and cell-based studies providing insight into signalling by the (epidermal growth factor receptor) EGFR through formation of asymmetric dimer is presented. Findings reveal the importance of dimer interface blockage in (mitogen-induced gene 6 or ERRFI1) MIG6-mediated inhibition.
