EGF receptor signaling stimulates SRC kinase phosphorylation of clathrin, influencing clathrin redistribution and EGF uptake
Article Abstract:
Signaling via the epidermal growth factor receptor (EGFR) stimulates SRC kinase activity. This occurs in response to EGF binding and results in CHC phosphorylation and clathrin redistribution to the cell periphery. This mechanism indicates that downstream receptor signaling has a direct effect upon the endocytic machinery. Specific inhibition or genetic deletion also delays endocytosis of EGF. These results suggest that clathrin-mediated membrane traffic can be regulated by EGFR signaling, defining a novel pathway in ligand-induced endocytosis for this receptor.

Regulation of clathrin assembly and trimerization defined using recombinant triskelion hubs
Article Abstract:
Part of the heavy chain fragment of clathrin was coexpressed in bacteria with a neuronal light chain to examine the nature and mechanism of clathrin assembly. Trimerization of the recombinant fragments resulted in a structure resembling the hub domain of the triskelion-shaped clathrin molecule, suggesting that clathrin light chains also play a role in clathrin coat formation. Deletion mutations showed that the hub domain mediates light chain binding in clathrin self-assembly.

Cargo-laden clathrin-coated pit (CCP)s is highly dynamic and could produce multiple vesicles in succession. It is proposed that CCP invagination, actin polymerization, and clathrin-coated vesicles (CCV) formation are highly coordinated for efficient endocytosis.
