Fibroblast growth factors, depolarization, and substratum interact in a combinatorial way to promote neuronal survival
Article Abstract:
Researchers used cultured chick ciliary ganglion neurons to examine the possibly interactive and combinatorial influence of trophic factors (acidic and basic fibroblast growth factor; FGF), depolarization and substrate, and to check whether neuronal survival in vivo may be influenced by the joint impact of multiple factors rather than by the impact of a single factor. The results show the possible significance of synergistic interactions between trophic elements and depolarizing stimuli.

Depolarization and laminin independently enable bFGF to promote neuronal survival through different second messenger pathways
Article Abstract:
Both laminin and depolarization can initiate cellular responsiveness to the neurotrophic factor basic fibroblast growth factor (bFGF) by acting through different second messenger pathways. The synergistic effect of bFGF and low levels of depolarization requires an inflow of calcium, since the calcium channel blocker PN200-110 prevented neuronal survival. The capability of laminin to enable bFGF to promote neuronal survival is obstructed by antibodies to beta-1-integrin.

Neuron-muscle contact changes presynaptic resting calcium set-point

Researchers used time-lapse video microscopy and fura-2 calcium studies to analyze contacts between presynaptic neurites of Helisoma motoneurons and muscle fibers delinked from identified partners in chemical synaptogenesis. They examined the function of retrograde signaling between target cells and their presynaptic partners during early cell-cell interactions. Calcium variations may be related to the early phases of synapse development.
