Identification of Aspergillus brlA response elements (BREs) by genetic selection in yeast
Article Abstract:
The role of brlA gene in transcriptional activation and in the identification of sites for BrlA polypeptide interaction was evaluated from a minimal yeast promoter fused with upstream sequences of a developmentally regulated Aspergillus nidulans rod A gene. The DNA fragment from the promoter region displayed mediation of brlA-dependent activation. Two other DNA fragments which responded to brlA and identified sites for BrlA polypeptide interaction were labelled as brlA response elements (BREs). The BRE-containing DNA fragments affected the developmental regulation of Aspergillus.

On the consistency of a physical mapping method to reconstruct a chromosome in vitro
Article Abstract:
Previous studies related to genetic mapping have failed to evaluate the statistical consistency of physical mapping methods in reconstructing chromosomes. Based on the assumption that a staircase of DNA fragments exists within large randomly generated recombinant DNA libraries, a model of physical mapping by binary fingerprinting of DNA fragments was developed. Through the model, and on the assumption that an epi-convergence of a physical mapping method exists, it can be established that such mapping techniques possess strong statistical consistency.

The use of simulated annealing in chromosome reconstruction experiments based on binary scoring
Article Abstract:
Simulated annealing, a method of combinatorial optimization, can be utilized to draw up the position of clones along a chromosome. The method assigns binary digital numbers to each clone depending on the existence of particular sequences in the clone as determined by its hybridization with synthetic oligonucleotide probes. The similarity of digital assignment or signature of each clone determines the overlap between clones which can, in turn, be used to map the chromosome.
