Insertional gene inactivation in a phototrophic sulphur bacterium: APS-reductase-deficient mutants of Chromatium vinosum
Article Abstract:
The cloning of the genes encoding the adenylylsulphate-forming enzyme adenosine-5'-phosphosulphate (APS) reductase from Chromatium vinosum strain D (DSM 180), a representative of the purple sulphur bacteria, and the construction of mutations in these genes via insertion of a kanamycin omega cartridge are described. The first insertional gene inactivation in purple sulphur bacteria was verified through southern hybridization and PCR analyses of the recombinants obtained. The absence of APS reductase from the mutants was noted, but this was found to have no significant effect on the sulphite-oxidizing ability of the cells under photolithoautotrophic growth conditions.

Truncation as a novel form of variation of the p50 gene in Mycoplasma hominis
Article Abstract:
Research was conducted to examine the existence of a variant of the p50, an adhesin molecule, in Mycoplasma hominis. To investigate the ubiquity of P50 in M hominis and to determine if the isolates, which did not react with the mAb, contain and express the gene encoding P50, the isolates were studied at the mRNA and DNA level. Results indicated that p50 gene sequences were present in all M hominis isolates that were examined.

Construction of a single-copy integration vector and its use to study gene expression in bacillus licheniformis
Article Abstract:
Bacillus licheniformis is used to produce peptide antibiotic bacitracin (Bt) alpha-amylase, beta-lactamase, alkaline protease and alkaline phosphatase. A versatile system to study gene expression in single copy is reported, integrated via a Campbell type system into the heterologous bts locus, comprising a promote-probe pTLH vector and Lac- mutant derivative of B. licheniformis.
