Isolation and characterization of ovochymase, a chymotrypsin-like protease released during Xenopus laevis egg activation
Article Abstract:
An easy two-step method that utilizes a soybean trypsin inhibitor-agarose affinity column helps isolate ovochymase, a 30-kDa chymotrypsin-like protease, from the perivitelline space of unactivated Xenopus eggs. The 30-kDa- and a 78-kDa protease present in the column eluant are separated by SDS-PAGE or gel filtration HPLC. The N-terminal amino aid sequence of ovochymase resembles those of other serine proteases. Substrate specificity and active site specific inhibition studies confirm that ovochymase is a member of the serine protease I family.
