Long G tails at both ends of human chromosomes suggest a C strand degradation mechanism for telomere shortening
Article Abstract:
The telomeric three prime overhangs of human chromosomes were analyzed by primer-extension-nick-translation (PENT) and nondenaturing hybridization in human primary fibroblasts, umbilical vein and leukocytes. PENT and nondenaturing hybridization indicated the presence of long 3' overhangs in cultured primary lung fibroblasts, umbilical vein endothelial cells and leukocytes. Furthermore, the three prime overhangs were produced by telomerase-independent mechanisms.

Nucleosomal organization of telomere-specific chromatin in rat
Article Abstract:
A study of the structure of telomere-specific chromatin in rat chromosomes is presented. The chromosomes of eukaryotes terminate with telomeres, special structures which have key roles in various phases of the cell cycle. Standard electrophoretic and sedimentation analyses of telomeres treated with nuclease showed that most telomeric DNA are organized into nucleoprotein structures which are indistingushable from nucleosomes.

Structure of the catalytic domain of human DOT1L, a non-SET domain nucleosomal histone methyltransferase
Article Abstract:
Crystallographic analyses of the catalytic domain of human Dot1 together with S-adenosyl-L-methionine (SAM) shows alpha-helical N-terminal domain and a central open alpha/beta structure, active site for SAM binding, and a lysine binding channel. Data indicate that the C-teminal catalytic domain consists of a positively charged stretch that is crucial for nucleosome binding and enzymatic activity.
