Mapping of 61 genes on the refined physical map of the chromosome of Thermus thermophilus HB27 and comparison of genome organization with that of T. thermophilus HB8
Article Abstract:
EcoRV and ClaI were used to refine the physical maps of the Thermus thermophilus HB27 chromosome and the large plasmid pTT27. A total of 49 restriction enzyme cleavage sites were identified on the HB27 chromosome. The location of 61 genes was determined on the maps. A significant similarity in the genome organization of the HB27 and HB8 strains was detected by comparing the refined HB27 maps and the HB8 chromosomal map. However, some genes were located in different regions of the genomes in the two strains.

Construction of a proline-producing mutant of the extremely thermophilic eubacterium Thermus thermophilus HB27
Article Abstract:
The method for the production of a proline-producing strain of Thermus thermophilus was investigated. A 3,4-dehydroproline-resistant mutant was developed through the introduction of mutations into the proB gene and subsequently, a mutant which cannot use proline for growth was isolated by mutagenizing the dehydroproline-resistant mutant. Seven mutants which do not have the ability to use L-proline for their growth were isolated.

Directed introduction of DNA cleavage sites to produce a high-resolution genetic and physical map of the Acinetobacter sp. strain and physical (BD413UE) chromosome
Article Abstract:
The physical and genetic map of the single, circular chromosome of Acinetobacter sp. strain ADP1 (BD413UE) was presented. Acinetobacter recipient strains were transformed by modified plasmid DNA to introduce novel endonuclease recognition sites in the chromosome. Southern hybridization techniques and transverse alternating field electrophoresis determined the positions of the recognition sites.
