New approaches to typing and identification of bacteria using the 16S-23S rDNA spacer region
Article Abstract:
The 16S-23S spacer region of the rRNA operon is useful in identifying microorganisms at the species and strain levels. The 16S rRNA gene is particularly popular as a method for identifying species, genera and families of bacteria. However, the use of the gene is time-consuming and costly. On the other hand, the 23S gene nor the 5S gene have not been widely used for phylogenetic purposes. Bacterial typing that utilizes both the 16S and 23S genes is proposed. The method is based on spacer-length variation and is less time-consuming and more cost-effective than the traditional method.

DNA sequence heterogeneity in the three copies of the long A6-S-23SrDNA spacer of Enterococcus faecalis isolates
Article Abstract:
Research was conducted to examine the possibility of intragenic heterogeneity between copies of the long intergenic 16S-23S rDNA spacer (LISR) region via specific amplification of the region from 21 Enterococcus faecalis isolates. The hybridization of the LISR to genomic DNA cleaved with I-CeuI and SmaI revealed three copies of the LISR. Results demonstrate that the transition between homogeneous and heterogeneous LISRs may be useful in the study of evolutionary mechanisms between E. faecalis isolates.

Typing of Stpahylococcus aureus strains by PCR-amplification of variable-length 16S-23S rDNA spacer regions: characterization of spacer sequences
Article Abstract:
A method for sequencing the genetic material of various strains of Staphylococcus aureus is described. The method involves hyperamplification of the spacer region C of its RNA operon to produce nucleotide sequences with lengths ranging from 906 to 1223 base pairs. Regions containing tRNa genes were also sequenced, some containing two tRNa genes. Ribotypes for penicillin- and methicillin-resistant strains were also described.
