Nuclear receptors in Sicily: all in the famiglia
Article Abstract:
There was a very interesting exchange of views at the EMBO workshop on the structure and function of nuclear receptors (NRs) which took place in May 1997 in Erice, Sicily. Research work relating to NRs, which act as molecular machines to transduce a hormone signal into a transcriptional response, has been going in a number of different directions. However, there has been a coming together recently of laboratories studying chromatin remodelling and those looking at NR cofactors.

BXR, an embryonic orphan nuclear receptor activated by a novel class of endogenous benzoate metabolites
Article Abstract:
BXR, a vertebrate orphan receptor, heterodimerizes with RXR and binds DNA sites with a variant thyroid hormone response element. BXR responds to low-molecular weight molecules with distinctive spectral patterns. The bioactivity-guided screen of embryonic extracts look different from other nuclear receptor ligands. Studies such as this can lead to a new understanding of physiologic homeostasis.

The histone acetylase PCAF is a nuclear receptor coactivator
Article Abstract:
The histone acetylase PCAF plays a bigger role in nuclear receptor function that it has been thought. Retinoid acid receptor and retinoid X receptor enlist the help of PCAF from mammalian cell extracts in a ligand-dependent manner. There is increased retinoid-responsive transcription when there is additional PCAF. The in vitro demonstration with PCAF also defines anew model of DNA binding.
