The Ink4a tumor suppressor gene product, p19(super Arf), interacts with MDM2 and neutralizes MDM2's inhibition of p53
Article Abstract:
Research was conducted to test whether the tumor suppressor gene product , p19(super Arf) hinders oncogenic transformation in primary cells and whether its role is abrogated when p53 is neutralized by viral oncoproteins and dominant-negative mutants. Cell cultures were monitored and maintained by calcium phosphate using the same amounts of DNA as in p53 degradation. Results showed high levels of genetic instability in p53 and that potent tumor suppressor activity resulted from the encoding of unrelated antioncogenic proteins with cooperating modes of action.

JNK targets p53 ubiquitination and degradation in nonstressed cells
Article Abstract:
The role of nonactive Jun-N (amino)-terminal kinase (JNK) in the regulation of stability of p53 has been studied. The quantity of p53-JNK complex related inversely to the level of p53 and ubiquitination of p53 was effectively obstructed by a peptide corresponding to the JNK binding site on p53. Also p53 without the JNK binding site showed a longer half-life than p53wt. It can be seen that JNK is an Mdm2-independent regulator of stability of p53 in unstressed cells.

Research has been conducted on the Mdm2 which is a ubiquitin ligase for the tumor suppressor protein p53. Results indicate that Mdm2 is conjugated with SUMO-1 (sumoylated) at Lys-446.
