GM-CSF and TNF-alpha cooperate in the generation of dendritic Langerhans cells
Article Abstract:
Antigen-presenting dendritic cells initiate a wide range of immune responses including organ transplant rejection. Though they originate from bone marrow, dendritic cells may have a maturation site in non-lymphoid tissue. They are found in many tissues such as mucosa or as Langerhans cells in the skin, but they are difficult to isolate and their growth process poorly understood. Granulocyte macrophage-colony stimulating factor with tumor necrosis factor-alpha is necessary for generating human Langerhans cells. This finding may make large samples of these cells available for research.

Article Abstract:
A mutant form of the tumor necrosis factor (TNF) molecule has been produced that may lack the toxicity that has so far prevented TNF from being used to treat cancer. Xaveer Van Ostade and colleagues used TNF's tendency to bind to two different cell-surface receptors, TNF-R55 and TNF-R75, to generate a TNF mutant with a 600-fold lower affinity for TNF-R75 but with full ability to bind to TNF-R55. However, tests for toxicity will be necessary before the mutant TNF's therapeutic usefulness can be properly assessed.
