Detection of Tn5-like sequences in kanamycin-resistant stream bacteria and environmental DNA
Article Abstract:
The utilization of growth colonies on media containing antibiotics for detecting kanamycin and neomycin resistance in the bacterial assemblage of a South Carolina stream is discussed. Kanamycin resistant, culturable stream bacteria were analyzed to determine if functional copies of neomycin phosphotransferase (npII) gene were abundantly present. Spatial differentiation was conducted in the relative abundances of npII in samples. The utility of DNA hybridization for assessing the abundance of genes in mixtures of nonculturable organisms wasdemonstrated.

Temporal variation in genetic diversity and structure of a lotic population of Burkholderia (Pseudomonas) cepacia
Article Abstract:
Multilocus enzyme electrophoresis is used to analyze the genetic structure and temporal patterns of genetic diversity in a population of Burholderia (Pseudomonas) cepacia. Allelic variation in seven structural gene loci is monitored at a single stream location for a maximum period of 32 days. Results show that these bacterial populations exhibit greater variability among sites than within a site over time.

Comparison of methods of DNA extraction from stream sediments
Article Abstract:
A comparative study of the efficiencies of three different techniques of DNA extraction from soils or sediments reveals that yield is higher in Ogram method than in other methods. The less time consuming Tsai method produces low quality DNA samples. The Jacobsen method produces pure DNA. The Ogram method produces DNA with a smaller proportions of euacterial DNA.
